The Melanoma Discovery: Bacteria in the Tumor Environment

Using confocal microscopy, researchers visualized Bartonella organisms nestled among VEGFC immunoreactivity in the dermal lymphatic vessels of melanoma tissue. In cell culture models, they confirmed that Bartonella henselae can invade the melanoma cell cytoplasm, triggering an increase in VEGFC and IL-8 production. This mechanistic pathway suggests that the bacteria may actively fuel cancer progression.
How Bartonella May Promote Tumor Progression
Bartonella is the confirmed cause of bacillary angiomatosis — it can directly induce vascular lesions via VEGF-dependent angiogenesis. In melanoma, a four-step pathway has been proposed:
Step 1 — Chronic Intracellular Infection: Bartonella colonizes the skin’s dermis, the same environment where melanoma thrives.
Step 2 — Entry into Melanoma Cells: The bacteria penetrate directly into the melanoma cell cytoplasm, placing the pathogen inside the cancer cell itself.
Step 3 — VEGFC and IL-8 Upregulation: Cultured cells show increased production of the pro-angiogenic factor VEGFC and the inflammatory cytokine IL-8.
Step 4 — Worsening Tumor Microenvironment: Elevated levels of VEGFC and IL-8 may accelerate tumor growth, enhance vessel recruitment, and promote metastatic spread.
The Critical Problem: Standard Tests Are Unreliable
Standard tests for Bartonella, including serology and conventional PCR, often miss evidence of active chronic infection.
